Radiosensitivity and cell cycle RADIOSENSIBILIDAD Y CICLO CELULAR

As we know, one of the main fields of activity of Radiology in Medicine is the treatment of tumors.

The target of any radiotherapy treatment is to kill the tumor cell with maximum respect the surrounding healthy tissue included in the radiation volume; for this, one of the main strategies that have radiologists is increasing the sensitivity of tumor cells to radiation by altering or inhibiting its repair mechanisms in the cell cycle.

1As all life is at risk of damage to their genetic material, we have some control points within the cell cycle in G1 phase (through the ATM, p53 and p21 proteins), in S phase (ATM Chk1/Chk2 , CDC25A , CD25C . BRCA1 , BRAC2 ), G2 phase start (ATM , Chk1 , Chk2 , CDC25A , CD25C , BRCA1 , BRAC2 ) and end of G2 ( ATR, Chk1 , CDC25A , CDC25C ) .

2The cell has several stops in the cycle where the checkpoint kinase 1 (Chk1) has been described as a central mediator and its activation not only phosphorylates CDC25 family leading to the arrest in G2 / M , but also Rad51 promotes DNA repair mediated double-strand break. Thus, Chk1 is crucial in the development of resistance to radiation, and as a result, inhibition of Chk1 can lead to death induced by preferential irradiation of the tumor cells. So in order to inhibit this control point has come a new model of AZD7762 control, a drug administered in conjunction with radiation and allows us to inhibit the Chk proteins, which in turn act on phosphatases, inhibiting repair and favoring damage accumulation in tumor cells with each successive division of the cells whose final results in the accumulation of large alterations in their genetic material that lead to tumor cell death. These proteins regulate CHKS good G1 / S transition step or G2 / M cell cycle.

One tumor which has become a central target of the radiation treatment is breast cancer, and in this study cited above, the high efficiency of co-administration of radiation therapy was shown together with the sensitizer in the treatment of cancer breast with mutations in the p53 protein.

As seen in the graph , in the combined treatment of AZD7762 and radiotherapy, induction of apoptotic death due to radiation is higher than if independently administered radiation or AZD7762 model control.

Furthermore, we have also noted that along with the critical role of these repair proteins, cell cycle within each of the phases has a greater or lesser degree of sensitivity to radiation, so that the phase when the G2- M radiosensitive, we aim to achieve the best therapeutic results with the least radiation dose, will be to ensure that the greatest number of tumor cells are in this phase at the time of applying radiation dose.

Como ya sabemos, uno de los principales campos de actuación de la Radiología en la Medicina es el tratamiento de los tumores.

El principal objetivo de todo tratamiento radioterápico es acabar con la célula tumoral respetando al máximo el tejido sano circundante incluido en el volumen de irradiación; para ello, una de las principales estrategias con las que cuentan los radiólogos consiste en el aumento de la sensibilidad de la célula tumoral a la radiación alterando o inhibiendo sus mecanismos de reparación en el ciclo celular.

1Como toda forma de vida está expuesta a sufrir daño en su material genético, contamos con unos puntos de control dentro del ciclo celular en Fase G1 (a través de las proteínas ATM, p53 y p21), en Fase S (ATM, Chk1/Chk2, CDC25a, CD25C. BRAC1, BRAC2), inicio fase G2 (ATM, Chk1, Chk2, CDC25A, CD25C, BRAC1, BRAC2) y final de G2 (ATR, Chk1, CDC25A, CDC25C).

2 La célula cuenta con varias paradas dentro del ciclo donde la quinasa de punto de control 1 (Chk1) ha sido descrita como un mediador central, y su activación no sólo fosforila la familia CDC25 que lleve al arresto en fase G2 / M, sino que también promueve al ADN Rad51 mediado por reparación de la rotura de doble cadena. Por lo tanto, Chk1 es crucial en el desarrollo de resistencia a la radiación, y como resultado, la inhibición de Chk1 puede conducir a una muerte inducida por la radiación preferencial de las células tumorales. De manera que con el objetivo de inhibir este punto de control ha salido un nuevo modelo de control AZD7762, una droga que se administra junto con la radiación y nos permite inhibir a las proteínas Chk, que a su vez, actúan sobre las fosfatasas, inhibiendo la reparación y favoreciendo la acumulación de daño en las células tumorales con cada división sucesiva de sus células, que tiene como resultado final la acumulación de grandes alteraciones en su material genético que conducen a la muerte de la célula tumoral. Estas proteínas Chks actúan regulando el paso bien G1/S o transiciones G2/M en el ciclo celular.

Uno de los tumores que se ha convertido en diana central del tratamiento con radioterapia, es el cáncer de mama, y en este estudio citado anteriormente, se demostró la alta eficacia de la administración conjunta de la radioterapia junto con este sensibilizador en el tratamiento del cáncer de mama con mutaciones en la proteína p53.

Como podemos observar en la gráfica, en el tratamiento combinado de AZD7762 y radioterapia, la inducción de muerte por apoptosis debida a la radiación es superior que si administramos de forma independiente radiación o modelo de control AZD7762.

Además, también tenemos que señalar que junto con el papel fundamental de estas proteínas reparadoras, dentro del ciclo celular cada una de las fases presenta un mayor o menor grado de sensibilidad a la radiación, de manera que al ser la Fase G2-M la más radiosensible, nuestro objetivo para conseguir los mejores resultados terapéuticos con la menor dosis de radiación, va a ser procurar que el mayor número de células tumorales se encuentren en esta fase en el momento de aplicar la dosis de radiación.
